Before a generic medicine ever reaches a pharmacy shelf, it passes through a defined sequence of scientific and regulatory testing. Here is what that sequence looks like.

The path a generic medicine travels from formulation to pharmacy shelf involves a defined sequence of scientific evaluation, each stage designed to answer a specific question about the product's identity, purity, performance, and stability. Understanding this sequence helps explain why regulatory approval of a generic is a meaningful scientific milestone rather than a formality.

The process begins with formulation development, in which chemists design the exact composition of the tablet, capsule, or liquid — selecting excipients that ensure the active ingredient remains stable, is released into the body appropriately, and can be manufactured consistently at scale. This stage includes analytical method development: the creation of validated laboratory techniques capable of precisely measuring the amount of active ingredient and detecting any impurities down to fractions of a percent.

Once a candidate formulation is ready, it proceeds to bioequivalence testing, described in detail elsewhere on this blog, in which the generic is compared directly against the branded reference product in human volunteers to confirm equivalent absorption into the bloodstream. In parallel, the manufacturer conducts impurity profiling and forced-degradation studies, deliberately exposing the formulation to heat, light, humidity, and pH extremes to identify every possible breakdown product the medicine could form under real-world storage conditions, and to confirm that none of those breakdown products exceed strict safety thresholds.

Stability testing runs alongside this work over an extended timeline — often two years or more — storing the product under controlled temperature and humidity conditions and testing it at regular intervals to establish an accurate, evidence-based shelf life. This is what allows a manufacturer to state with confidence, rather than assumption, exactly how long a medicine will remain within specification after it is packaged.

Every batch that comes off the production line during this process, and every commercial batch manufactured after approval, is tested against a release specification covering identity, potency, dissolution rate, and impurity levels before it is permitted to leave the facility. Samples from each batch are also retained for a defined period, so that if any quality question arises after distribution, the exact batch in question can be re-examined directly rather than relying on records alone.

Only after formulation development, bioequivalence confirmation, impurity and stability studies, and a validated manufacturing process have all been reviewed and accepted by a regulatory authority does a generic medicine receive marketing approval. It is a demanding, multi-year scientific process — and it is the reason a properly approved generic can be trusted to perform exactly as its regulatory dossier says it will.
